question on having the PCB manufactured

I have a general question.

Is there an initial flashing with a programmer needed before flashing via USB works - in case the device gets produced based on the schematics provided? 

You wrote that USB flashing is possible with empty chip in #11 . This mean it works "fresh from fab" without initial flashing of anything?
